CEM will present a solid-phase peptide synthesis (SPPS) process that eliminates solvent-intensive washing steps during each amino acid addition cycle. A significant breakthrough involves removing a volatile Fmoc deprotection base through bulk evaporation at elevated temperatures, preventing condensation on vessel surfaces with directed headspace gas flushing. Demonstrated at both research and production scales, this process showed no impact on product quality, even when applied to challenging sequences (up to 89 amino acids in length). The outcome is an exceptionally fast, high-purity, scalable process with a substantial waste reduction (up to 95%), requiring only 10–15% of the standard amount of base used. This transformation in SPPS signifies a significant improvement in peptide manufacturing process efficiency, potentially expanding access to peptide-based therapeutics.
